    Review of Current Neuroimaging Studies of the Effects of Prenatal Drug Exposure: Brain Structure and Function

    Neuroimaging tools have provided novel methods for understanding the impact of prenatal drug exposure on brain structure and function and its relation to development and behavior. Information gained from neuroimaging studies allows for the investigation of how prenatal drug exposure alters the typical developmental trajectory. The current prevalence and characteristics of prenatal drug exposure and its implications for vulnerable periods of brain development are reviewed. Structural and functional neuroimaging methods are introduced with examples of how study results from prenatal alcohol, cocaine, marijuana, and tobacco exposure further our understanding of the neurodevelopment impact of prenatal drug exposure. Prenatal drug neuroimaging studies have advanced our understanding of mechanisms and functional deficits associated with prenatal drug exposure. Studies have identified brain circuits associated with the default mode network, inhibitory control, and working memory that show differences in function as a result of prenatal drug exposure. The information gained from studies of prenatal drug exposure on brain structure and function can be used to make connections between animal models and human studies of prenatal drug exposure, identify biomarkers of documented effects of prenatal drug exposure on behavior, and inform prevention and intervention programs for young children

    Computing evolutionary distinctiveness indices in large scale analysis

    We present optimal linear time algorithms for computing the Shapley values and 'heightened evolutionary distinctiveness' (HED) scores for the set of taxa in a phylogenetic tree. We demonstrate the efficiency of these new algorithms by applying them to a set of 10,000 reasonable 5139-species mammal trees. This is the first time these indices have been computed on such a large taxon and we contrast our finding with an ad-hoc index for mammals, fair proportion (FP), used by the Zoological Society of London's EDGE programme. Our empirical results follow expectations. In particular, the Shapley values are very strongly correlated with the FP scores, but provide a higher weight to the few monotremes that comprise the sister to all other mammals. We also find that the HED score, which measures a species' unique contribution to future subsets as function of the probability that close relatives will go extinct, is very sensitive to the estimated probabilities. When they are low, HED scores are less than FP scores, and approach the simple measure of a species' age. Deviations (like the Solendon genus of the West Indies) occur when sister species are both at high risk of extinction and their clade roots deep in the tree. Conversely, when endangered species have higher probabilities of being lost, HED scores can be greater than FP scores and species like the African elephant Loxondonta africana, the two solendons and the thumbless bat Furipterus horrens can move up the rankings. We suggest that conservation attention be applied to such species that carry genetic responsibility for imperiled close relatives. We also briefly discuss extensions of Shapley values and HED scores that are possible with the algorithms presented here

    An exploration of infant and toddler unstructured outdoor play

    Unstructured outdoor play is important for children\u27s development. The present study examined infants’ and toddlers’ physical and social unstructured outdoor play behaviors within childcare centers. Children\u27s outdoor play behaviors were video recorded at two centers (A&B) and assessed using a modified version of the Observational System for Recording Physical Activity in Children in Preschool (OSRAC-P). Children in this study primarily took part in active play (56.7%) and engaged in play without any type of prompting from a teacher (91.2%). There was a significant interaction between the effect of center and location on physically active play (p \u3c 0.001). Children at Center B were most active in open play areas (77.6%), while children at Center A were most active in gross motor play areas (72.2%). In sum, the outdoor play environment influences infants’ and toddlers’ physical and social play behaviors; however, more research is needed to determine the optimal environment for development

    Examining Configural, Metric, and Scalar Invariance of the Pain Catastrophizing Scale In Native American and Non-Hispanic White Adults In the Oklahoma Study of Native American Pain Risk (OK-SNAP)

    Introduction: Native Americans (NAs) have a higher prevalence of chronic pain than other US racial/ethnic groups, but the mechanisms contributing to this pain disparity are under-researched. Pain catastrophizing is one of the most important psychosocial predictors of negative pain outcomes, and the Pain Catastrophizing Scale (PCS) has been established as a reliable and valid measure of the pain catastrophizing construct. However, before the PCS can be used to study pain risk in NAs, it is prudent to first determine whether the established 3-factor structure of the PCS also holds true for NAs. Methods: The current study examined the measurement (configural, metric, and scalar) invariance of the PCS in a healthy, pain-free sample of 138 NA and 144 non-Hispanic white (NHW) participants. Results: Results suggest that the previously established 3-factor solution fits for both groups (configural invariance) and that the factor loadings were equivalent across groups (metric invariance). Scalar invariance was also established, except for 1 minor scalar difference in a single threshold for item 3 (suggesting NHWs were more likely to respond with a 4 on that item than NAs). Discussion: Results provide additional evidence for the psychometric properties of the PCS and suggest it can be used to study pain catastrophizing in healthy, pain-free NA samples

    Bilateral Cochlear Implantation for Children in Nagasaki, Japan

    Objectives: The number of patients with bilateral cochlear implant (CI) has gradually increased as patients and/or parents recognize its effectiveness. The purpose of this report is to evaluate the efficacy of 29 bilateral CI out of 169 pediatric CI users, who received auditory-verbal/oral habilitation at our hearing center. Methods: We evaluated the audiological abilities 29 Japanese children with bilateral CIs including wearing threshold, word recognition score, speech discrimination score at 1 m from front speaker (SP), 1 m from second CI side SP, speech discrimination score under the noise (S/N ratio=80 dB sound pressure level [SPL]/70 dB SPL, 10 dB) at 1 m from front SP, word recognition score under the noise (S/N ratio=80 dB SPL/70 dB SPL, 10 dB) at 1 m from front SP. Results: Binaural hearing using bilateral CI is better than first CI in all speech understanding tests. Especially, there were significant differences between the results of first CI and bilateral CI on SDS at 70 dB SPL (P=0.02), SDS at 1 m from second CI side SP at 60 dB SPL (P=0.02), word recognition score (WRS) at 1 m from second CI side SP at 60 dB SPL (P=0.02), speech discrimination score (SDS) at 1 m from front SP under the noise (S/N=80/70; P=0.01) and WRS at 1 m from front SP under the noise (S/N=80/70; P=0.002). At every age, a second CI is very effective. However, the results of under 9 years old were better than of over 9 years old on the mean SDS under the noise (S/N=80/70) on second CI (P=0.04). About use of a hearing aid (HA) in their opposite side of first CI, on the WRS and SDS under the noise, there were significant differences between the group of over 3 years and the group of under 10 months of HA non user before second CI. Conclusion: These results may show important binaural effectiveness such as binaural summation and head shadow effect. Bilateral CI is very useful medical intervention for many children with severe-to-profound hearing loss in Japan as well as elsewhere

    Quantitative proteomics identify Tenascin-C as a promoter of lung cancer progression and contributor to a signature prognostic of patient survival

    The extracellular microenvironment is an integral component of normal and diseased tissues that is poorly understood owing to its complexity. To investigate the contribution of the microenvironment to lung fibrosis and adenocarcinoma progression, two pathologies characterized by excessive stromal expansion, we used mouse models to characterize the extracellular matrix (ECM) composition of normal lung, fibrotic lung, lung tumors, and metastases. Using quantitative proteomics, we identified and assayed the abundance of 113 ECM proteins, which revealed robust ECM protein signatures unique to fibrosis, primary tumors, or metastases. These analyses indicated significantly increased abundance of several S100 proteins, including Fibronectin and Tenascin-C (Tnc), in primary lung tumors and associated lymph node metastases compared with normal tissue. We further showed that Tnc expression is repressed by the transcription factor Nkx2-1, a well-established suppressor of metastatic progression. We found that increasing the levels of Tnc, via CRISPR-mediated transcriptional activation of the endogenous gene, enhanced the metastatic dissemination of lung adenocarcinoma cells. Interrogation of human cancer gene expression data revealed that high TNC expression correlates with worse prognosis for lung adenocarcinoma, and that a three-gene expression signature comprising TNC, S100A10, and S100A11 is a robust predictor of patient survival independent of age, sex, smoking history, and mutational load. Our findings suggest that the poorly understood ECM composition of the fibrotic and tumor microenvironment is an underexplored source of diagnostic markers and potential therapeutic targets for cancer patients

    Fluency expresses implicit knowledge of tonal symmetry

    The purposes of the present study were twofold. First, we sought to establish whether tonal symmetry produces processing fluency. Second, we sought to explore whether symmetry and chunk strength express themselves differently in fluency, as an indication of different mechanisms being involved for sub- and supra-finite state processing. Across two experiments, participants were asked to listen to and memorize artificial poetry showing a mirror symmetry (an inversion, i.e., a type of cross serial dependency); after this training phase, people completed a four-choice RT task in which they were presented with new artificial poetry. Participants were required to identify the stimulus displayed. We found that symmetry sped up responding to the second half of strings, indicating a fluency effect. Furthermore, there was a dissociation between fluency effects arising from symmetry vs. chunk strength, with stronger fluency effects for symmetry rather than chunks in the second half of strings. Taken together, we conjecture a divide between finite state and supra-finite state mechanisms in learning grammatical sequences
